This kit uses a simple alkaline lysis approach to release DNA, making it ideal for routine genotyping, pathogen detection, and DNA barcoding.
The extraction process involves a two-step method of cell lysis and neutralisation, performed in a single 0.2ml PCR tube. Samples are incubated with a dilute alkaline solution and then neutralised, completing DNA extraction in 10–30 minutes depending on tissue type.
This one-tube format minimises handling, reduces contamination risk, and allows for easy scaling from a few samples to hundreds per day.
The kit’s alkaline lysis solution targets DNA-rich tissues that are low in PCR inhibitors. It is suitable for extracting DNA from animal tissues, invertebrates, parasite blood meals, zooplanktonic eggs, young plant leaves, pollen, and fungal cultures.
While efficient for PCR applications, it is not recommended for obtaining large quantities of unfragmented genomic DNA or samples with high levels of PCR inhibitors.
This kit contains two reagents: Alkaline Lysis Solution (25mM NaOH, pH 12) and Neutralising Buffer (100mM Tris-HCl, 0.5mM EDTA, pH 8), supplied in two 30ml bottles providing up to 400 reactions.
Routine storage is at room temperature, with care to minimise air exposure. For longer-term storage, the pH of the alkaline solution should be checked prior to use. The kit is shipped at room temperature.
